Determination of Metal Ion Content in Animal Feed Using Atomic Absorption Spectrophotometry

Key words: atomic absorption spectrophotometer; animal feed; heavy metal; analysis; method

This procedure outlines the determination of metal ion content in animal feed using an atomic absorption spectrophotometer (AA1800). The method involves sample preparation, digestion, and measurement under controlled conditions to ensure accurate results. It is suitable for quantifying elements such as calcium (Ca), copper (Cu), iron (Fe), magnesium (Mg), potassium (K), manganese (Mn), sodium (Na), and zinc (Zn).

The process begins with the selection of appropriate laboratory equipment, including a platinum, quartz, or porcelain crucible, a muffle furnace, and a hot plate. Reagents like hydrochloric acid (HCl), cerium nitrate, and barium chloride are used for sample digestion and calibration. Standard solutions are prepared to construct calibration curves, which are essential for quantitative analysis.

Sample preparation includes drying, ashing, and dissolution in hydrochloric acid. After digestion, the solution is filtered and diluted before being analyzed. Blank solutions are prepared alongside samples to account for background interference. The absorbance values obtained from the instrument are corrected using blank measurements, and the concentrations of metals are calculated based on the calibration curves.

For each element, specific wavelengths are used for measurement: Cu at 324.8 nm, Fe at 248.3 nm, Mn at 279.5 nm, Zn at 213.8 nm, Ca at 422.6 nm, K at 766.5 nm, Mg at 285.2 nm, and Na at 589.6 nm. The results are expressed in mg/kg or g/kg, depending on the concentration range, and adjusted according to the provided table for accuracy.

This method ensures reliable detection of heavy metals in animal feed, supporting quality control and safety assessments in the agricultural industry.

Content Range Correction Factor
5mg/kg - 10mg/kg 0.1mg/kg
10mg/kg - 100mg/kg 1mg/kg
100mg/kg - 1g/kg 10mg/kg
1g/kg - 10g/kg 100mg/kg
10g/kg - 100g/kg 1g/kg
